Pregabalin (Lyrica)

Mechanism of action

Anticonvulsant, anxiolytic and analgesic medication.

Mechanism of action still not fully understood. 

Binding to presynaptic voltage-gated calcium channels and modulation of the release of several excitatory neurotransmitters is probably responsible for the antiseizure and antinociceptive effects of pregabalin.

Pharmacokinetics

Onset of action: 1.5 hours

Half-life of elimination: 6.3 hours

Metabolism: Less than 2 % of pregabalin is metabolized

Elimination: Kidneys (as unchanged drug)

Medicinal forms

Tablets – 25 mg, 50 mg, 75 mg, 100 mg, 150 mg, 200 mg, 225 mg, 300 mg

Side-effects

Dizziness, somnolence, headache

Increased appetite

Mood changes, confusion, irritability, decreased libido

Vomiting, nausea, diarrhoea

Ataxia, tremor, dysarthria, amnesia, attention disorders

Diplopia, blurred vision

Vertigo
